ABSTRACT

<p><b>OBJECTIVE</b>To study the effects of xijiao dihuang decoction on the expressions of Bcl-2 and caspase-3 protein positive cells in brain tissue, TNF-alpha and IL-6 in blood after acute intracerebral hemorrhage in rat.</p><p><b>METHOD</b>The experimental ICH model was established by injection of non-heparinized autologous arterial blood into the right basal ganglia. Immunohistochemistry and radioimmunity methods were used to detect expressions of Bcl-2 and caspase-3 protein positive cells in brain tissue, TNF-alpha and IL-6 in blood.</p><p><b>RESULT</b>After ICH, caspase-3 protein positive cells in brain tissue, TNF-alpha and IL-6 in blood increased obviously in the model group, caspase-3 protein positive cells, TNF-alpha and IL-6 were significantly reduced by treatment with Xijiao Dihuang decoction, on the contrary, Bcl-2 protein positive cells in brain tissue increased markedly.</p><p><b>CONCLUSION</b>Xijiao Dihuang decoction can pretect nerve cell by reducing the expression of caspase-3, TNF-alpha and IL-6, increasing the expression of Bcl-2 after ICH in hemorrhagic brain damage.</p>

ABSTRACT

Objective To investigate the changes of distribution and expression of NADPH\|d positive neurons and Fos like immunoreactive(Fos\|LI) neurons in the rats medullary visceral zone(MVZ) following different time course after global cerebral ischemia reprefussion. Method Fifty adult SD rats were divided into three groups,normal control group( n =2),sham operation group( n =12) and ischemia group( n =36).The 4 brain vessel of ischemia group rats were occluded for 30?min,then reperfused for 2?h,4?h,8?h,12?h,24?h and 48?h respectively.The sections of medulla oblongata were divided into three series.The first and second series were reacted with NADPH\|d histochemical and anti\|Fos immunohistochemical methods respectively,and the third series were double stained with the other two methods mentioned above. Results NADPH\|d neurons and Fos\|LI neurons were distributed in bilateral MVZ(in NTS and VLM predominately).The number of NADPH\|d neurons and Fos\|LI neurons of ischemia group rats were significantly increased than that of control group at 2\|12?h.Approximately 15\|20 percent NADPH\|d/Fos\|LI double labeled neurons were found in MVZ.Conclusion\ The results indicated that NADPH\|d neurons and non\|NADPH\|d neurons could sensitively respond to brain ischemia reperfsion and might associate with regulation of stress and defence responese.

ABSTRACT

Objective To explore the relationship between the expression of c fos gene and delayed neuron death in perinatal hypoxia ischamia encephalopathy (HIE). Methods Terminal deo xynuleotidyl transferase mediated dUTP nick end labeling (TUNEL), immuno histochemistry andreverse transcription PCR(RT PCR) were used to detect the apoptosis and transcription and translation of c fos gene in the hippocampus following HIE. Results Positive signal of apoptosis from CA1 to CA4 sections were observed in experimental hippocampi. Marked signal appeared earlier and longer in CA1 section than in the other sections[CA1: 1 h: (14.6?2.3),24 h: (51?6),72 h: (17.4?0.3);CA4:1 h :1.3?1.6),24 h: (47?8),72 h: (21.6?0.6) apoptosis cell number/ mm 2 ],compared with the sham group of CA1 section P

ABSTRACT

Objective To explore the magnetic resonance imging tracking of superparamagnetic iron oxid(SPIO) labeled embryonic neural stem cells (NSCs) transplantation into corpus striatum of the focal cerebral ischemia(FCI) rats and the effects of learning and memory abilities.Methods NSCs of embryonic rat were cultured in vitro and labeled with Fe2O3-mediated by poly-L lysine (PLL), with Prussian blue staining to evaluate the labeling rate and Trypan blue staining to observe cell viability of NSCs. The rats were randomly divided into groups A (normal control),B (normal labeled NSCs implanted),C (cerebral ischemia),D( labeled NSCs implanted),E (unlabeled NSCs implanted)and F (inactivated labeled NSCs implanted).The FCI models were made in groups C, D, E, F.The labeled NSCs or unlabeled NSCs or inactivated labeled NSCs were stereotaxically injected into the left corpus striatum of groups B,D,E,F respectively. The abilities of learning and memory were measured by the Y-type electronical maze in groups A, C, D, E after 3 d,7 d,2,3,4 weeks of transplantation respectively. MRI scanning was performed to monitor the NSCs in groups B, D, F at corresponding time. After MRI, the rats were killed and undergone Prussian blue staining of the brain histological section to observe the distribution of implanted NSCs.Results The labeling efficiency rate and cell viability of Fe2O3-PLL labeled NSCs were almost 100% and 95% respectively. ABSTRACT

Objective The proliferation and differentiation of neural stem cells in the subventricular zone(SVZ) and subgranular zone(SGZ) have been observed in normal old rats or treated by middle cerebral artery occluded(MCAO) and sham operation respectively. Methods Old rats were used to make models of middle cerebral artery occluded(MCAO).By BrdU pulse labeling and immunohistochemical single staining,the change of the number of BrdU positive cells was observed in SVZ and SGZ of normal rats or cerebral ischemia rats after MCAO 3d,7d,14d,21d,28d and sham operation respectively.By BrdU accumulating label and immunohistochemical double staining,BrdU/NeuN and BrdU/GFAP positive cells were determined in SVZ and SGZ of aged rats after MCAO 14d. Results BrdU positive cells were found in bilateral SVZ and SGZ of all groups above mentioned.Compared with normal and sham groups,BrdU positive cells of SVZ and SGZ in cerebral ischemia rats were strikingly increased.BrdU positive cells reached the peaks in SVZ at 7d and in SGZ at 14d after MCAO.At 14d after MCAO,BrdU/NeuN(0.98%) or BrdU/GFAP(12.56%) double labled cells were observed in SVZ,but no double labled cell was observed in SGZ.Conclusion The focal cerebral ischemia markedly stimulated proliferation of neural stem cells in SVZ and SGZ in old rats,and some proliferative cells in SVZ can differentiate into neurons or glial cells.
